В настоящее время ВД КРС распространена практически во всех странах мира с интенсивным ведением животноводства. Особая актуальность проблемы заключается в большом экономическом ущербе, который складывается из снижения удоя во время болезни, гибели молодняка от серозной пневмонии, недополучения привесов живой массы у молодняка, потери продуктивности и воспроизводства животных и, как следствие, абортов и мертворождений, рождения нежизнеспособных телят, а также проведения профилактических, карантинных и ликвидационных мероприятий. Важным звеном в недопущении распространения вирусной диареи крупного рогатого скота остается оперативное проведение лабораторных исследований. Одним из самых технологичных методов диагностики является полимеразная цепная реакция в режиме реального времени.</p></sec><sec><title>Методика</title><p>Методика. Оценку диагностической значимости тест-системы ОТ-ПЦР VetMAX BVDV Screening (Thermo Fisher) проводили по показателям чувствительности, специфичности и прецизионности в условиях повторяемости и воспроизводимости. Для определения повторяемости проводили исследование 5-ти образцов культуры клеток ПТ-80, зараженной эталонным штаммом «Орегон 24» и 7 образцов культуры клеток ПТ-80, зараженной изолятом «Ресса», в десятикратных разведениях от 10–1 до 10-7, одним оператором в трех параллельных исследованиях на одном оборудовании. Для определения воспроизводимости и чувствительности — тех же образцов тремя операторами в разные дни на одном оборудовании. Для определения специфичности проводили исследования 3 образцов, не содержащих вирус вирусной диареи — аденовирус КРС 1 типа, штамм Bovina — 10, вирус ринотрахеита КРС, штамм «Оренбург» и вирус парагриппа, 3 штамм ЗКСМ.</p></sec><sec><title>Результаты</title><p>Результаты. После математической обработки результатов постановки ПЦР для оценки воспроизводимости получены следующие результаты: коэффициент вариации (СV) для тест-системы VetMAX BVDV Screening (Thermo Fisher) составлял 1,0–4,0%; коэффициент вариации (CV) при оценке повторяемости — 1–3%. Специфичность тест-систем VetMAX BVDV Screening (Thermo Fisher) составляла 100%. Тест-система VetMAX BVDV Screening (Thermo Fisher) чувствительна в отношении обнаружения генома вируса вирусной диареи КРС. Так, при постановке ПЦР проб изолята «Ресса» в разведении 10–7 значения Сt определились на 38,18–39,24 циклах, эталонного вируса ВД «Орегон 24» в разведении 10–5 — на 37,85–39,45 циклах амплификации.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Relevance</title><p>Relevance. Currently, BVD is widespread in almost all countries of the world with intensive livestock farming. The special relevance of the problem lies in the large economic damage that consists of a decrease in milk yield during the disease, the death of young animals from serous pneumonia, the loss of live weight gain in young animals, the loss of productivity and reproduction of animals and, as a result, abortions and stillbirths, the birth of non-viable calves, as well as preventive, quarantine and liquidation measures. An important link in preventing the spread of viral diarrhea in cattle remains the rapid conduct of laboratory research. One of the most technologically advanced diagnostic methods is real-time polymerase chain reaction.</p></sec><sec><title>Methods</title><p>Methods. The diagnostic significance of the VetMAX BVDV Screening (Thermo Fisher) RT-PCR test system was assessed by sensitivity, specificity, and precision in conditions of repeatability and reproducibility. To determine repeatability, 5 samples of PT-80 cell culture infected with the reference strain "Oregon 24" and 7 samples of PT-80 cell culture infected with Ressa isolate were studied in tenfold dilutions from 10-1 to 10-7, by one operator in three parallel studies on the same equipment. To determine reproducibility and sensitivity, 5 samples of PT-80 cell culture infected with the reference strain "Oregon 24" and 7 samples of PT-80 cell culture infected with «Ressa» isolate were studied in ten-fold dilutions from 10-1 to 10-7 , by three operators on different days on the same equipment.</p><p>To determine the specificity, studies were conducted on 3 samples that did not contain the virus of viral diarrhea — bovine adenovirus type 1 strain Bovina — 10, rhinotracheitis virus of cattle strain "Orenburg" and parainfluenza virus 3 strain ZKSM.</p></sec><sec><title>Results</title><p>Results. After mathematical processing of the results of PCR formulation for the assessment of reproducibility, the following results were obtained: the coefficient of variation (CV) for the VetMAX BVDV Screening (Thermo Fisher) test system was from 1.0–4.0 %; the Coefficient of variation (CV) for the assessment of repeatability was 1–3 %. The specificity of the test systems VetMAX BVDV Screening (Thermo Fisher) was 100%. The VetMAX BVDV Screening (Thermo Fisher) test system is sensitive to detecting the genome of the bovine viral diarrhea virus.
