Инсулиноподобный фактор роста 1 (insulin-like growth factor-1, IGF-1) вовлечен in vivo в регуляцию мейоза, в связи с чем активно изучается как потенциальный регулятор качества ооцитов с целью повышения эффективности биотехнологий, направленных на получение эмбрионов in vitro. В настоящей работе исследовали влияние IGF-1 в культуре ооцитов коров с точки зрения их созревания in vitro и эмбрионального развития после искусственной активации, а также качества полученных эмбрионов.</p></sec><sec><title>Методы</title><p>Методы. Выделенные из овариальных фолликулов ооциты культивировали в отсутствие (контроль) и присутствии различных концентраций (5, 10, 20, 40 и 80 нг/мл) IGF-1. Затем созревшие ооциты искусственно активировали и культивировали в течение 7 дней для эмбрионального развития.</p></sec><sec><title>Результаты</title><p>Результаты. IGF-1 не повлиял на созревание ооцитов, а также на их дробление после партеногенетической активации и число ядер в 7-дневных эмбрионах. В то же время обнаружен зависящий от концентрации эффект IGF-1 на развитие ооцитов до стадии бластоцисты. В контроле выход бластоцист составлял 17,3±1,6%. Присутствие в среде созревания IGF-1 в концентрациях 5, 10 и 20 нг/мл повышало этот показатель, соответственно, до 29,9±4,5, 30,7±3,6 и 29,6±2,3% (p &lt; 0,05). Следовательно, в этих концентрациях IGF-1 может быть рекомендован для in vitro созревания ооцитов коров. </p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Relevance</title><p>Relevance. Insulin-like growth factor-1 (IGF-1) is involved in the regulation of meiosis in vivo and is therefore actively investigated as a potential regulator of oocyte quality aimed at improving the efficiency of in vitro embryo production biotechnologies. In the present study, we examined the effect of IGF-1 during bovine oocyte culture in terms of their in vitro maturation, embryonic development after artificial activation, and the quality of the resulting embryos.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Oocytes isolated from ovarian follicles were cultured either in the absence (control) or presence of various concentrations of IGF-1 (5, 10, 20, 40, and 80 ng/ml). Matured oocytes were then artificially activated and cultured for 7 days to support embryonic development.</p></sec><sec><title>Results</title><p>Results. IGF-1 had no effect on oocyte maturation, cleavage, or the number of nuclei in blastocyst. At the same time, a concentration-dependent effect of IGF-1 on the development of oocytes to the blastocyst stage was observed. In the control group, the blastocyst yield was 17.3±1.6%. The presence of IGF-1 in the maturation medium at concentrations of 5, 10, and 20 ng/ml increased this rate to 29.9±4.5, 30.7±3.6, and 29.6±2.3%, respectively (p &lt; 0.05).
