Developing technological approaches aimed at increasing the adaptive capacity of in vitro-produced grape plants to ex vitro conditions

Abstract
Relevance. The use of biotechnological methods can lead to innovative and efficient ways to propagate and health improvement agricultural crops, including grapes. The adaptation of plants from in vitro to ex vitro conditions is one of the most important stages of clonal micropropagation technology, which directly determines the efficiency of the process.
Methods. A study was conducted to investigate technological possibilities for increasing the adaptive capacity of in vitro-produced grape plants to ex vitro conditions. The rootstocks Gravesac 11, Gravesac 12, Ruggeri 140 and Fercal clone 242 were used as research objects. Clonal micropropagation of grapes was carried out in the laminar flow cabinet AHC-4D1. The phytotron LiA-2 was used to adapt plants to ex vitro conditions. An experimental batch of in vitro plants of the Fercal clone 242 rootstock underwent adaptation and further growth on ALVM installations.
Results. The operating instructions for the LiA-2 phytotron, which include seven sequential schemes, were developed during the experiment. 65 plants of the rootstocks Gravesac 11, Gravesac 12, Ruggeri 140 and Fercal clone 242, adapted to ex vitro conditions, were obtained during the experiment. The results indicated differentiated plant viability, which is due to the high level of lethality at the initial stage of the work. Testing of a pilot batch of 146 plants of Fercal clone 242 rootstock, obtained in vitro, adapted to ex vitro conditions, and further grown on AOBM, demonstrated a high survival rate of 75% in soil conditions.
